Abstract—In this paper, a process for large-scale purification of high purity C-phycocyanin (C-PC) has been set up. Spirulina platensis dried powder was incubated with 1 mg/ml lysozyme and then ruptured by using high pressure homogenizer. The crude extract was precipitated by 50% saturation ammonium sulfate, and purified by hydrophobic interaction chromatography using Phenyl Sepharose 6 FF, ion exchange chromatography using DEAE Sepharose FF and gel filtration chromatography using Sephacryl S-100 HR. The final recovery of C-PC was 42.03% with purity ratio (A620/A280) of 5.32.
